Abstract

A LLA caracterizada pela presença do rearranjo com o gene de linhagem mista MLL possui frequência em todos os estágios de idade, sendo considerado um marcador de obscuro prognóstico. A partir disto, o MLL possui uma diversidade de mais de 80 genes com capacidade de translocar-se. Dentre todos os genes que podem formar o rearranjo com o MLL, existem aqueles que possuem uma maior frequência e que em sua maioria, desempenham semelhança no fenótipo de LLA, de onde surge a indagação do motivo de tamanha semelhança, sendo estes genes tão distantes e distintos funcionalmente e molecularmente. O presente estudo buscou analisar a semelhança entre os genes MLL; AF4; AF6; AF9 e AF10, através da metodologia filogenética de máxima parcimônia. Foi observado uma grande distinção entre os genes analisados, como já esperado, contudo, os genes apresentaram maior quantidade de sítios conservados em seu estado selvagem, do que em seu estado translocado. O motivo da similaridade fenotípica e o método exato de atuação não está claro, sendo sugerido que as características similares podem estar relacionadas pela aquisição de um domínio N – terminal funcional (vinda do gene parceiro) ao qual é ausente na proteína do MLL selvagem, facilitando a interação com moléculas adjacentes e ligação ao DNA. Contudo, mais estudos são necessários para melhor compreensão da dinâmica entre os genes envolvidos no rearranjo MLL. 7
