Aplicação de tanase parcialmente purificada em teste de digestão in vitro de animais monogástricos / Application of partially purified tannase in vitro testing of monogastric animals digestion

Resumo

Tanases são enzimas que hidrolisam as moléculas de taninos hidrolisáveis. Sua utilização, em alimentos, contribui para remover os efeitos indesejáveis dos taninos. O objetivo do presente trabalho foi produzir e aplicar tanase de Aspergillus niger URM 7131 em teste de digestão in vitro de animais monogástricos. A produção enzimática ocorreu sob fermentação submersa. Após este período o extrato enzimático bruto passou por uma purificação parcial por meio de membranas de ultrafiltração. Foi avaliada a atividade enzimática e conteúdo proteico. Após purificação parcial verificou-se o efeito do pH e temperatura na atividade e estabilidade enzimática, e então procedeu-se a aplicação no teste de digestão in vitro. Na purificação parcial foi possível obter um fator de purificação de 3,8. A tanase apresentou um ótimo a 40ºC, foi estável entre 30 a 60 ºC, retendo entre 67,1 a 100%. Teve sua atividade ótima no pH 4, sendo também mais estável neste pH após 180 minutos. A tanase de reteve 159,3% após a simulação total digestão in vitro. Esses resultados sugerem que a enzima pode ser empregada como aditivo de rações animais. Verifica-se a necessidade de novoAditivo de rações, Aspergillus niger, Tanino Acil Hidrolase, Ultrafiltração.s estudos in vivo visando verificar a eficiência catalítica da tanase obtida de A. niger URM7131.
